Project description:
The aim of this project is to analyze pathological changes in DNA methylation in gingival cells from patients with periodontitis, identify the influence of Porphyromonas gingialis on DNA methylation in these cells and characterize its biological consequences. Studies performed within this project will not only improve our understanding of this disease, but can also facilitate identification of new therapeutic targets, constituting a foundation for testing new drug classes in the treatment of periodontal disease in the future.  
 
Key responsibilities: 
  1. In vitro and in situ studies of global and promoter-specific changes in DNA methylation in gingival fibroblasts (GFs), periodontal ligament fibroblasts (PLFs) and in total gingival tissue. 
  2. Determination of the role of P. gingivalis infection and inflammation in promoter-specific DNA methylation in gingival cells. 
  3. Functional studies of biological consequences of altered DNA methylation in gingival cells. 

Profile of candidates/requirements:

  1. MSc degree (or equivalent) in biotechnology, biochemistry or any related field of life sciences. 
  2. Strong experience in primary eukaryotic cell culture and in basic cell biology techniques (ELISA, Western Blot, qPCR) evidenced by results presented in MSc thesis or publication. 
  3. 3. Experience in culturing anaerobic bacteria. 
  4. Experience in epigenetic studies (DNA and histone modification, regulation of epigenetic enzymes) would be an advantage. 
  5. Fluency in written and spoken English. 
  6. Good skills in work organization, planning and keeping experimental track record. 
  7. High level of independence, determination and reliability, willingness to work outside of standard office hours. 
  8. Ability to work in a team.
